Characterization of the cytokines secretion by mouse mesangial cell line MES-13 and primary murine tubular epithelial cells

Introduction: Chronic kidney disease (CKD) is irreversible when it goes through G3b phase. Inflammation contributes to the progression of CKD by inducing the release of cytokines and the increased production and activity of adhesion molecules, which together results in immune responses and fibrosis in kidney. In the present study, we investigate the cytokines secreted by different type of renal cells and whether their secretions can be affected by gamma-aminobutyric acid (GABA), which was reported to have a beneficial effect on renal function in nephrectomized rats by inhibiting fibrosis and atrophy in tubuli. Materials and Methods: Mouse tubular epithelial cells (mTEC) were isolated from 6-weeks to 8-weeks C57BL/6 mice. mTEC or MES-13 cells, a mouse mesangial cell line, were pre-treated with or without GABA for 1 hr, and stimulated with LPS for 24 hr. Gene expression of cox-2 in MES-13 were measured by real-time PCR. The cytokine levels of monocyte chemoattractant protein (MCP)-1, IL-6 and TGF-β were also measured. Data were analyzed by Student t test. Results: The cytokines secretions of glomerular mesangial cell line MES-13 cells and tubular epithelial cells were different. MES-13 cells secreted higher MCP-1 but mTEC cells secreted little. LPS stimulation increased MCP-1 secretions by both cells. MES-13 cells did not secrete IL-6 at all, but mTEC secreted little IL-6 and increased 10-fold under LPS stimulation. Both MES-13 cells and mTEC could secrete TGF-β without LPS stimulation. GABA pre-treatment decreased MCP-1 and TGF-β secretions from LPS-stimulated MES-13 cells. The cox-2 expression in LPS-stimulated MES-13 significantly increased, but GABA pre-treatment suppressed this cox-2 expression. Conclusions: These data showed the cytokines secretion profiles of glomerular mesangial cell line MES-13 cells and tubular epithelial cells were different. GABA might suppress proinflammatory cytokines secretion in renal cells.
