The approach for SDGs based on cultivation of Suizenji-nori, as a raw material of the ingredient, Sacran, for cosmetics

Introduction
Suizenji-nori (Aphanothece sacrum) is a freshwater unicellular cyanobacteria endemic to Japan. Originally, Suizenji-nori was found in the river basin from Aso caldera, where is well known volcano in Japan. This species is used to grow wild in Lake Ezu in Kumamoto City, where is in the downstream of the river basin. However, this species had been decreased and completely disappeared in this lake. Nowadays, this species is only being preserved through aquaculture in a few districts in Fukuoka and Kumamoto, so that this is now listed as an endangered species in Japan.
Sacran, a polysaccharide sulfate extracted from the extracellular matrix of Suizenji-nori, is one of the world's largest natural polymers (16-27 MDa). Because of its unique structure, in which there are 11 kinds of saccharides, the characteristic abilities of this ingredient are very high water retentivity and barrier effect of skin by thin film-forming on human skin. The demand for this natural ingredinet is increasing in the cosmetics industry.
However, the productivity of Suizenji-nori is limited because it can only grow in the pure water from Aso caldera. Therefore, it is very the urgent issue to preserve the aquatic environment and to protect the species and to rise the productivity of Suizenji-nori. Because of this situation, we attempted to revitalizing aquaculture and industry based on the environmental conservation. In this study, we report our efforts of the aquaculture of Suizenji-nori and the industrial utilization of Sacran to achieve the social construction based on the SDGs. We are aiming at developing natural ingredient from the cultivation of endangered cyanobacteria to the cosmetics ingredient based on preserving natural environment.
Trial of suizenji-nori cultivation
In Kumamoto City area, there are a lot of fountain water from Aso caldera. However, the quality of the water is getting worse because of house waste and the quantity of the water is decreased because of the usage in the city. So, we are trying to use the fountain water in Aso caldera. Although there are a lot of natural fountain water souses, whose quality are good in this area, the temperature of the souses is lower than that in Kumamoto City for the cultivation of suizenji-nori. We have been controlling the temperature with greenhouse (not to use heat generator) and searching the source, higher temperature.
Utilization of Sacran
Nowadays, we have several kinds of chemicals which will affect for the skin, such as pollution environments. For these two years, COVID-19 has been going around all over the world, so people wear masks of chemical material. Human skin is exposed these kinds of unfavorable conditions. Sacran extracted from Suizenji-nori will make unique film on human skin in application. That means that we will have barrier effect with Sacran. We have made evaluations of barrier effects on the skin with various parameters, such as water retaining rate, TEWL, reactive oxygen species and so on, before and after application of skin lotion containing Sacran.
Discussion & Conclusion
Utilizing fountain water gushing out naturally, we have made trials to cultivate Suizenji-nori, which grows a few places using and listed as endangered species in Japan. We have searched suitable some fountain sources in Aso caldera area. And controlling temperature of these fountain water without using heat generators, namely naturally.
We have already known that the ingredient extracted from Suizenji-nori, Sacran, has very unique polysaccharide for cosmetics. Cosmetics containing Sacran show barrier effects on human skin, protection against unfavorable chemicals, by forming film on skin. Growing naturally endangered species yields very lucrative ingredient for cosmetics, especially skin care, protecting unfavorable chemicals. This way is the approach of SDGs to sustain natural condition and utilize this for cosmetics industry. Namely, this study will suggest the sustainable development of the unique material from the nature.
