Reduction of Melanin in Melanocytes, Anti-inflammatory Effect on Macrophages, Soothing Effect on Mast Cells and AGEs clearance in bovine serum cells of Moutan Extract

Moutan, a symbol of good fortune in China, it was used by wealthy families in ancient China, and its root bark is a common material in Traditional Chinese Medicine. At present, the research on moutan bark is mostly focused on the field of medicine. It has anti-bacterial, anti-inflammatory and anti-allergic effects through oral solution, and it can also inhibit 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) from inducing Parkinson's symptoms, or improve motor coordination by reducing neuroinflammation. However, there were few studies on the roles of moutan bark in cosmetics, and therefore the studied of efficacy on soothing, whitening, anti-inflammatory and inhibition of non-enzymatic glycosylation of related cells by the extract of moutan root bark is our target. The moutan bark was heated by microwave and extracted by ultrasonic distillation with 50wt% ethanol/water. After decolorization with D101, the bark root extract was obtained after concentration. Different concentrations of the extract were used for in vitro cell experiments. The results showed that the activity of mast cells and macrophages was affected when the concentration of moutan root bark extract was greater than 0.625%, while the activity of melanocytes was inhibited when the concentration was greater than 0.156%. Further studies showed that the extraction of moutan root bark (0.3125%, 0.625%) could reduce the degranulation response of mast cells after C48/80 stimulation, and the degranulation rates were 31.69±3.52% and 25.86±0.85% respectively. It is speculated that the extract of moutan root bark can alleviate the occurrence of type I hypersensitivity by reducing the degranulation rate of mast cells. In addition, Rac1, Cdc42, GP100 and TRP1 are four genes that produce melanin-related proteins in melanocytes. In our study, 0.156% moutan extract can reduce the expression levels of these four genes, and thus reduce the melanin content in melanocytes. In addition, low concentration of moutan root bark extract can reduce the expression of inflammatory factors IL-6 and TNF-α and play an anti-inflammatory role, on the contrary, high concentration of moutan root bark extract can reduce the release of NO. Finally, moutan root bark extract can effectively remove AGEs in bovine serum protein. These results proved that the extract of moutan root bark had soothing, whitening, anti-inflammatory and inhibition of non-enzymatic glycoylation effect, and can be used as the effective raw material in cosmetics products.
